ACE-031 is a recombinant ActRIIB-Fc fusion protein investigated for its ability to inhibit myostatin and related TGF-β ligands that regulate skeletal muscle growth. It has been studied in preclinical models and early human clinical trials for its potential effects on lean body mass, muscle strength and body composition.

Research Overview
About ACE-031 1mg
ACE-031 is a soluble activin receptor type IIB (ActRIIB) fusion protein that acts as a ligand trap. Rather than stimulating muscle growth directly, it binds circulating proteins such as myostatin and activins before they reach their cellular receptors. Researchers have investigated this mechanism as a potential strategy for increasing skeletal muscle mass in conditions associated with muscle wasting. Although early studies demonstrated increases in lean body mass, clinical development was discontinued following vascular-related adverse events.
How It Works in Research
ACE-031 functions as a soluble decoy receptor derived from activin receptor type IIB. Once administered, it binds myostatin, activin A, GDF-11 and several related ligands belonging to the transforming growth factor-beta (TGF-β) superfamily. These proteins normally suppress muscle growth by activating ActRIIB receptors on skeletal muscle cells. By preventing this interaction, ACE-031 reduces inhibitory signalling and allows researchers to investigate increased muscle protein synthesis, muscle fibre growth and improvements in lean body mass. Unlike anabolic steroids, ACE-031 does not activate androgen receptors, and unlike growth hormone secretagogues it does not rely on the GH/IGF-1 axis. Instead it targets one of the body's primary regulators of muscle growth. Early clinical studies demonstrated measurable increases in lean body mass, but development was halted after vascular-related adverse events were observed, meaning long-term efficacy and safety remain uncertain.
Research Applications
• Muscle hypertrophy • Sarcopenia • Muscular dystrophy • Cachexia • Rehabilitation research • Body composition

Key Research Characteristics
Recombinant ActRIIB-Fc fusion protein
Myostatin/Activin pathway inhibitor Long-acting biologic Investigated in muscle wasting disorders Early clinical and extensive preclinical research
Potential Benefits in Research
Preclinical and early clinical studies investigated ACE-031 for increasing lean body mass, improving muscle strength, enhancing recovery from muscle wasting conditions and improving overall body composition. Researchers also explored its use in Duchenne muscular dystrophy and other neuromuscular disorders where preservation of muscle mass may be beneficial. These findings remain investigational and should not be interpreted as established therapeutic outcomes.
Safety Profile
Early human studies reported epistaxis (nosebleeds), telangiectasia (small dilated blood vessels), headache and mild injection-site reactions. These vascular findings contributed to discontinuation of clinical development. Long-term human safety is unknown because larger clinical trials were not completed.

REFERENCE RECONSTITUTION INFORMATION
Evidence Status: Early clinical and preclinical research. Mechanism: Myostatin and activin ligand trap. Reconstitution: Add 2.0 mL bacteriostatic water to one 1 mg vial to produce a concentration of 0.5 mg/mL. Published studies used weight-based investigational dosing rather than fixed-dose research vials.
Always follow sterile preparation techniques when handling research peptides.
General Preparation
- Using a sterile syringe, slowly add 2.0 mL bacteriostatic water down the inside wall of the vial. Allow the powder to dissolve naturally, then gently swirl until the solution is clear. Do not shake vigorously. Refrigerate immediately after reconstitution.
